Core losses analysis of the LCL filter inductor for SiC-based inverter
The ability of SiC devices to switch at high speed allows increasing significantly the power density in both converters and passive components, reducing their required size. To mitigate harmonic injection form inverters into the grid, in order to comply with power quality standards, an accurate filter design is required. Given its excellent performance, an LCL filter is the configuration most suitable in grid-connected power converters. Several parameters must be considered when designing an effective LCL filter, and the inverter-side inductor assumes a special importance because of its relevance to suppress high frequency harmonic content at the inverter side. One of the most relevant issues to be considered in the process of designing the LCL filter is the evaluation of core losses in the inverter-side inductor, which will determine the final temperature of the inductor. This paper analyses the core losses of the inverter-side inductor of an LCL filter. The proposed method is based on the computation of the current harmonics generated by the inverter and on Steinmetzâs empirical equation. As a result, core losses calculated taking into account several carrier and sideband harmonics show good agreement with the experimental values. When current harmonics are estimated by simulation, as it is done in the proposed design procedure, results are less accurate, but precise enough for a design procedure
Morphine leads to global genome changes in H3K27me3 levels via a Polycomb Repressive Complex 2 (PRC2) self-regulatory mechanism in mESCs
Background: Environmentally induced epigenetic changes can lead to health problems or disease, but the mechanisms involved remain unclear. Morphine can pass through the placental barrier leading to abnormal embryo development. However, the mechanism by which morphine causes these effects and how they sometimes persist into adulthood is not well known. To unravel the morphine-induced chromatin alterations involved in aberrant embryo development, we explored the role of the H3K27me3/PRC2 repressive complex in gene expression and its transmission across cellular generations in response to morphine.
Results: Using mouse embryonic stem cells as a model system, we found that chronic morphine treatment induces a global downregulation of the histone modification H3K27me3. Conversely, ChIP-Seq showed a remarkable increase in H3K27me3 levels at specific genomic sites, particularly promoters, disrupting selective target genes related to embryo development, cell cycle and metabolism. Through a self-regulatory mechanism, morphine downregulated the transcription of PRC2 components responsible for H3K27me3 by enriching high H3K27me3 levels at the promoter region. Downregulation of PRC2 components persisted for at least 48 h (4 cell cycles) following morphine removal, though promoter H3K27me3 levels returned to control levels.
Conclusions: Morphine induces targeting of the PRC2 complex to selected promoters, including those of PRC2 components, leading to characteristic changes in gene expression and a global reduction in H3K27me3. Following morphine removal, enhanced promoter H3K27me3 levels revert to normal sooner than global H3K27me3 or PRC2 component transcript levels. We suggest that H3K27me3 is involved in initiating morphine-induced changes in gene expression, but not in their maintenance.This study was supported by grants from the Spanish Health Department ISCIII (DTS 18/00142) and University of the Basque Country. Quantum Conductance in Semimetallic Bismuth Nanocontacts
Electronic transport properties of bismuth nanocontacts are analyzed by means
of a low temperature scanning tunneling microscope. The subquantum steps
observed in the conductance versus elongation curves give evidence of atomic
rearrangements in the contact. The underlying quantum nature of the conductance
reveals itself through peaks in the conductance histograms. The shape of the
conductance curves at 77 K is well described by a simple gliding mechanism for
the contact evolution during elongation. The strikingly different behaviour at
4 K suggests a charge carrier transition from light to heavy ones as the

Efficacy and safety of native versus pegylated Escherichia coli asparaginase for treatment of adults with high-risk, Philadelphia chromosome-negative acute lymphoblastic leukemia
Native or pegylated (PEG) asparaginase (ASP) are commonly used in treatment of acute lymphoblastic leukemia (ALL), but have been scarcely compared in the same trial in adult patients. Native vs. PEG-ASP administered according to availability in each center were prospectively evaluated in adults with high-risk ALL. Ninety-one patients received native ASP and 35 PEG-ASP in induction. No significant differences were observed in complete remission, minimal residual disease levels after induction and after consolidation, disease-free survival, and overall survival. No significant differences in grades 3â4 toxicity were observed in the induction period, although a trend for higher hepatic toxicity was observed in patients receiving PEG-ASP. Chemotherapy or allogeneic transplantation in high-risk Philadelphia chromosomeânegative adult lymphoblastic leukemia
The need for allogeneic hematopoietic stem cell transplantation (allo-HSCT) in adults with Philadelphia chromosomeânegative (Phâ) acute lymphoblastic leukemia (ALL) with high-risk (HR) features and adequate measurable residual disease (MRD) clearance remains unclear. The aim of the ALL-HR-11 trial was to evaluate the outcomes of HR Phâ adult ALL patients following chemotherapy or allo-HSCT administered based on end-induction and consolidation MRD levels. Patients aged 15 to 60 years with HR-ALL in complete response (CR) and MRD levels (centrally assessed by 8-color flow cytometry) <0.1% after induction and <0.01% after early consolidation were assigned to receive delayed consolidation and maintenance therapy up to 2 years in CR. The remaining patients were allocated to allo-HSCT. CR was attained in 315/348 patients (91%), with MRD <0.1% after induction in 220/289 patients (76%). By intention-to-treat, 218 patients were assigned to chemotherapy and 106 to allo-HSCT. The 5-year (±95% confidence interval) cumulative incidence of relapse (CIR), overall survival (OS), and event-free survival probabilities for the whole series were 43% ± 7%, 49% ± 7%, and 40% ± 6%, respectively, with CIR and OS rates of 45% ± 8% and 59% ± 9% for patients assigned to chemotherapy and of 40% ± 12% and 38% ± 11% for those assigned to allo-HSCT, respectively. Our results show that avoiding allo-HSCT does not hamper the outcomes of HR Phâ adult ALL patients up to 60 years with adequate MRD response after induction and consolidation. Better postremission alternative therapies are especially needed for patients with poor MRD clearance
Adverse prognostic impact of complex karyotype (â„3 cytogenetic alterations) in adult T-cell acute lymphoblastic leukemia (T-ALL)
© 2021 The Author(s).The potential prognostic value of conventional karyotyping in adult T-cell acute lymphoblastic leukemia (T-ALL) remains an open question. We hypothesized that a modified cytogenetic classification, based on the number and type of cytogenetic abnormalities, would allow the identification of high-risk adult T-ALL patients. Complex karyotype defined by the presence of â„3 cytogenetic alterations identified T-ALL patients with poor prognosis in this study. Karyotypes with â„3 abnormalities accounted for 16 % (22/139) of all evaluable karyotypes, corresponding to the largest poor prognosis cytogenetic subgroup of T-ALL identified so far. Patients carrying karyotypes with â„3 cytogenetic alterations showed a significantly inferior response to therapy, and a poor outcome in terms of event-free survival (EFS), overall survival (OS) and cumulative incidence of relapse (CIR), independently of other baseline characteristics and the end-induction minimal residual disease (MRD) level.
